剩余污泥中蛋白质的资源化利用是目前国内外研究的热点,污泥预处理则是实现污泥中蛋白质释放的重要途径。为了进一步提高剩余污泥中蛋白质的溶出效果,本文选取热碱预处理,超声-碱联合预处理,溶菌酶预处理对污泥进行溶胞,以蛋白质提取浓度为主要指标进行参数优化,并利用等电点法对粗提取蛋白进行纯化回收。结果表明：热碱预处理(pH=13.0,温度140oC,时间1.5 h)溶胞效果最好,在最优条件下蛋白质提取浓度达到2062.98 mg.L-1,纯化回收率可达62.42 %。实验所得处理条件可为蛋白质的资源化利用提供更精确的技术参考。
The resource utilization of protein in residual sludge is a research hotspot at home and abroad, and sludge pretreatment is an important way to realize protein release from sludge. In order to further improve the dissolution efficiency of protein in residual sludge, this paper selected thermal-alkali pretreatment, ultrasonic-alkali pretreatment and lysozyme pretreatment as the method of disintegration in sludge, and optimized the parameters with the protein extraction concentration as the main index. The isoelectric point method was also used to purify and recover crude protein. The results showed that the best cell disintegration effect was achieved by thermal-alkali pretreatment (pH=13, temperature 140oC, time 1.5 h). Protein extraction concentration reached at 2,062.98 mg?L-1 and the purification recovery rate reached at 62.42% under the optimal conditions. The obtained pretreatment conditions can provide a more precise technology reference for protein resource utilization.
